Summary
Delicious cake is something that is seen in the game Portal. In Portal, cake is offered as a reason for the main character to complete ridiculous puzzles. The game does not really explain why the player wants this cake, but one /b/tard has rationalized it as such:
Portal never actually mentions delicious cake or the player's imperative to eat it, but it does directly say that the cake is a lie.
Delicious cake puzzle threads come out when the OP puts out an image featuring the main character, the delicious cake (often, but not always, a lie), and some obstacles to overcome. Perhaps the most common solution is to go 3D and walk around the obstacles. This is a great way to piss off the OP.

Origins
It was popularized by Portal where cake is the apparent reward for doing tests. But there really is no cake.
The most likely origin of the phrase "delicious cake was an old word used by 4channers to refer to child porn. The saying predates Portal - Some estimate its origins to be from early 2003.
